Getting from Mykonos to Kylini
Kylini is a small port town on the western coast of the Peloponnese and the truth is that almost nobody travels to Kylini itself. It exists as the main departure point for ferries to Zakynthos and Kefalonia, which means if you are heading here you are almost certainly using it as a connection point to one of those islands rather than as a destination in its own right.
Take the ferry
There is no direct ferry from Mykonos to Kylini. The practical route is to take the ferry from Mykonos to Piraeus, which takes around 2.5 to 5 hours depending on the vessel, and then travel by land from Athens to Kylini. The drive from Athens to Kylini takes around 3 to 3.5 hours heading west on the E65 motorway through the Peloponnese. From Kylini port, ferries to Zakynthos take around 1.5 hours and ferries to Kefalonia take around 1 hour to Poros port.
What about flying?
There is no airport in Kylini. If you are using Kylini as a connection point to Zakynthos or Kefalonia, flying directly to either of those islands via Athens is a faster option than routing through Kylini. The journey from Mykonos to Athens by air takes under an hour and onward flights to both islands are available from Athens Airport.
Your best option
If Kylini is your final destination, take the ferry to Piraeus and drive across. If you are using Kylini to connect to Zakynthos or Kefalonia, consider whether flying directly to those islands via Athens might be quicker and simpler than the Kylini ferry route. Both options are valid depending on whether you are travelling with a vehicle or as a foot passenger.
